To test for protein in milk, xanthoproteic reaction test should be carried out by the student. The procedure includes:
- To the milk solution add a few drops of concentrated nitric acid
- A white precipitate forms, which turns yellow on heating.
- Cool the contents and add a few drops of ammonia solution.
- The colour of the precipitate deepens to yellow indicating presence of protein in the cow milk.
To test for carbohydrate in milk, a test for complex sugar should be carried out by the student. The procedure includes:
- Add milk solution and hydrochloric acid into a test tube.
- Place the test tube inside a boiling water-bath.
- The acid hydrolyses the complex sugar to simple sugars.
- Fehling's test is then carried out on the simple sugars that are formed which are glucose and galactose.
- An orange-red precipitate indicates a positive result.
