Enterobacter aerogenes culture from an EMB plate should not be Gram stained because ideally Gram negative bacteria stain pink in the process but because of being cultured in EMB plate, Enterobacter aerogenes already have a purplish color on them.

EMB plate contains the Eosin Methylene Blue agar. It is a test used to distinguish Gram negative bacteria from a sample of species, as only they can survive on the plate. Eosin Y and methylene blue are pH indicator dyes. Their combination forms a dark purple precipitate at low pH.

Gram staining is the test used to classify the bacterial species into two classes: Gram positive and Gram negative. The gram positive bacteria yield a purple color in the test while the gram negative bacteria yield a pink color.
